Kelce ultimately deleted the post following its unexpected resurgence.
The tweet in question read: "Happy Easter to all! Shoutout to Jesus for takin one for the team... haha".
Kelce appeared amused by his past social media activity during the podcast discussion.
"So ridiculous," he said when reflecting on the post. "My favourite part of it is the 'Ha ha' at the end."
The Chiefs star explained his thinking behind the tweet's lighthearted tone.
"Listen, I wanted to make sure everybody knew I was having a good time joking about this," Kelce told listeners.
Jason Kelce offered a humorous defence of his brother's Easter message during the podcast.
"Listen. We don't give Jesus enough shout-outs for taking one for the team if we're all honest," the retired Philadelphia Eagles centre said.

Travis appeared to appreciate his brother's support, responding: "He's the best leader out there."
Viewers of the April 23 episode also noticed Kelce's distinctly tanned appearance whilst welcoming NFL Hall of Famer Joe Thomas to the show.
This sparked speculation that the 35-year-old tight end has been enjoying a well-deserved offseason holiday.

The podcast background differed from Kelce's usual Kansas City mansion setup, further fuelling vacation rumours.
Kelce and girlfriend Taylor Swift recently enjoyed a ski trip to Montana, though the couple have maintained a low profile.
They have only been spotted in New York once over the past couple of months.
Reports have suggested that Kelce and Swift are attempting to 'tone down' their public personas in the wake of the Chiefs' Super Bowl defeat back in February.
It has even been suggested that the couple will limit their public appearances so the NFL star can focus on his day job.
Some expected Kelce to walk away after the Chiefs were thumped by the Philadelphia Eagles back in February.
However, the 35-year-old has since confirmed he will keep playing the sport he loves - having been left disappointed by his performances last term.
